A Winfrey spokeswoman on Monday issued a statement saying Lee died at her Milwaukee home on November 22.
After meeting Vernon Winfrey, Vernita welcomed her first child in rural MS at the age of 18.
Winfrey's relationship with her mother changed for the better over the years, and Lee was occasionally spotted on Winfrey's daytime talk show, which ran on ABC from 1986 to 2011.
Her brother Jeffrey died of AIDS in 1989, and Patricia, who died in 2003.
Winfrey has always been outspoken about the tumultuous relationship she had with her mother when she was young.
Previously, the media mogul said she learned a surprising piece of information about herself online, that she was the very first African-American billionaire, and the first African-American woman as well.
"I loved her the way that she loves people now", Lee said of their bond.
"I suddenly land in a place that's completely foreign to me. I don't really even know my mother", Oprah said of the childhood move, Newsweek reported. "I was asking the question ... what is a mother?"
